iPSC generation

We generate transgene-free iPSCs using Sendai viral vectors, providing a safe and non-integrating alternative to traditional reprogramming methods. All iPSC lines are issued after thorough characterization (see below), confirmation of clearance of reprogramming vectors and mycoplasma testing.

Our standard iPSC generation package includes three different fully characterized clones per line. Additional clones can be provided upon request.

iPSC characterization

Our standard iPSC characterization includes:

  • Immunostaining for pluripotency markers (OCT4 and SSEA4)
  • Pluripotency analysis using directed differentiation to all three germ layers
  • Molecular karyotyping of three expanded clones
    • At least one clone without major genomic alterations will be provided
  • STR fingerprinting to confirm cell line identity

iPSC registration

We facilitate registration of iPSC lines at the Human Pluripotent Stem Cell Registry and can optionally assist with publication in Stem Cell Research. A hPSCreg® certificate shows ethical provenance and verified pluripotency based on standardized criteria. It is possible to opt-out from registration.

Time line and costs

The generation of fully characterized iPSC lines takes about 6 months. Timelines may vary depending on project requirements and current service availability. Our standard full-service package is priced at €6000 per donor (internal price). For project intake, availability, or special requests, please contact us at iPSC@amsterdamumc.nl
